How badly do I need an axle /engine alignment? - (see picture)

In my very long quest for fixing a vibration issue that occurs in my allroad 2.7T between 2700-3000RPM at light load, third gear, I today realized that I have failed to pay any attention to the fact that the engine-subframe alignment holes are not totally in line. On the passenger side, the upper hole is approx. 1 cm off compared to the lower two which are aligned, ref. picture. On the driver side, all three holes are straight in line.
Could this be a cause to shudder and vibration?
Your thoughts on the matter would be much appreciated.
