So I have a huge flat spot in my tire (from it being flat)...
this flat spot is no ordainary flat spot, the car was shaking so much it was practically undrivable over 40 mph. Will it eventually work it's way out? Tires are 225/40/18 Yoko ES100's. I got a small dent in my wheel and over the weekend it slowly leaked air til it was pretty flat, so it could have been sitting like that for 2 days. Bend wasn't bad enough that it couldn't hold air, just a slow leak, so I pumped it back up and drove to get it swapped with my spare. Wondering if I could still use this tire on another rim.

If the bend was bad enough that air was leaking...
it's bad enough to cause some nasty vibrations. A flat-spotted wheel could also cause vibrations...I'm just not really sure if 2 days is enough to permanently damage the tire.
I'd give it a try on another rim. If it still vibrates, then toss the tire.
