I would say that depends on which one is a better starting point for your trip. The Stuttgart area is nice, but so is the Munich area. I know the Stuttgart area better. There's the AUTO & TECHNIK MUSEUM SINSHEIM nearby. Pretty cool museum. It's where one of the decommissioned Concords ended up and you can go inside. Also, Neckarsulm is the birth place of all RS vehicles and the R8. It's where Audi Sport (formerly Quattro Gmbh) is located, so you might be able to tour their factory.

At Neckarsulm, they usually schedule only one USA delivery a day. For the factory tour, you can choose to tour the R8 assembly building (~1 hour) or the regular tour (~2-2.5 hours). These tours are personal tours, not a group tour. Your delivery person will give you a walking tour of the factory. The regular will take you through 3 buildings and see 6s, 7s, avants etc. coming down one assembly line. Where workers are assembling the cars, you are literally standing right next to them. Unfortunately no photos or videos allowed. This tour makes the European Delivery a great experience.
No museum, but they did have some Audi 100 models displayed and at another area, 4-6 super custom RS and S cars that you can sit in.
Someone else who picked their car up at Ingolstadt can describe their experience there.
Why did I choose Neckarsulm for my delivery? My original delivery was scheduled for Ingolastadt but I wanted to make a change to my order but it was too late so Stephanie (previous coordinator) canceled my original order and had me re-order my car immediately (mid July) to get the earliest production date and delay my delivery only one month. Brad then took over and to get the delivery date I wanted (early Oct.), 2-3 weeks sooner than at Ingolstadt, Neckarsulm was available. Didn't want late October or November (2106) because they would put winter tires on then vs all season tires. I actually took delivery about 3 weeks after my car was built, thanks to Brad.

My delivery specialist at Neckarsulm, Martin, spoke English very well. If you just went there for the tour, you will be in a group and you get a earphone.
They make exceptions for 1 US customer per day since another US customer took delivery the same day as me. That day, they only delivered 80 cars because half the room was set-up for a dinner function for that evening. They normally deliver about 120 cars a day there. It would be interesting to find out how many US deliveries (Ingolstadt & Neckarsulm) they do a year.
Maybe you can find out by contacting Brad after you get your Comm# for your car order. He didn't seem to have a problem arranging the delivery date I requested.

Well, if you are going in one shot it's a 2+ hour drive, so you would take the A9 and A6. Plenty of stretches with no speed limit, but obviously the car won't be broken it yet, so you shouldn't quite go ***** out. If you want to take your time or even stop in between, then you can drive the surface streets along A9 and A6 and meander through the towns along the route.

We had no issues keeping her below 4K RPMs while still hitting 120mph+.
However, on a previous drive from Munich to Neckarsulm we took the B16 to B25 to the A7. The "B" roads are not unlimited so no worries about break-in and was still a great drive.
Amazed at the hop farms in area. Acres and acres of glorious hops...

I ended up visiting four auto museums during my trip - BMW, Audi, Porsche, and Mercedes. Alas, the Audi museum was the least impressive. I also did factory tours at Audi and Porsche.
I couldn't fit Neckarsulm into my schedule. Hope to return one day to see Neckarsulm and the Technik museum.
