Default Could a sensor or the ECU cause misfires?

I noticed the signal (checked with o-scope) to the coilpack drops when a load is placed on the motor. If the ecu was defective could it not send the signal to the coilpack?
The misfire only happens in cylinder #6 and I have tried all the usual stuff, coilpack, sparkplug, maf, output amplifier, temp sensor, fuel injector/cleaner and checked the harness from the output amplifier to coilpack.
