Eh, it's easy enough to fill a drilled hole if you want to go back.

Just tap it with a tapered pipe tap and put in a pipe plug using plenty of sealant. Drilling while the manifold is ON the engine would just be plain crazy as the chips would get into the manifold and be sucked into the engine.
