Yes you should open the bleeder screw
It prevents contamination from being forced back upstream into the ABS components. That is standard procedure for all brake jobs at all our service centers

Rather than opening the bleed screw when pushing the pistons back, I bleed the caliper before pushing the pistons back.
I do this is for the same reason -- to avoid pushing contaminated brake fluid backwards into the master cylinder and/or ABS components. Since I track the car, this procedure removes old over-heated brake fluid and gets fresh fluid into the caliper every time I swap pads. Also, it seems every time I bleed brakes I remove at least one tiny bubble from somewhere, which firms up the pedal.
